What Is TB 500?
TB-500 is a synthetic peptide derived from a protein that all humans naturally have in their bodies. Specifically, this natural protein, which occurs in cells, is called Thymosin Beta 4. Notably, this protein may have healing properties. Interestingly, its peptide was first discovered in a lab in the 1960s. Initially, it was used for horses to prevent adhesions from forming. Furthermore, it has the potential to help speed up injury recovery. Typically, some injuries and wounds could take several months or longer to heal on their own. However, its peptide can significantly reduce that healing time.

Scientific Information
A veterinary preparation known as TB-500, containing a synthetic version of the naturally occurring peptide LKKTETQ, has recently emerged. Specifically, the peptide segment (17) LKKTETQ(23) serves as the active site within the protein thymosin β(4), which is responsible for actin binding, cell migration, and wound healing. In particular, the key ingredient is the peptide LKKTETQ, with artificial acetylation of the N-terminus. It is further claimed that this peptide promotes endothelial cell differentiation, angiogenesis in dermal tissues, keratinocyte migration, collagen deposition, and decreased inflammation. Consequently, to control the misuse of this compound in equine sports, a method to identify its prior use in horses is required. In response to this need, this study describes a method for the simultaneous detection of N-acetylated LKKTETQ and its metabolites in equine urine and plasma samples.
The possible metabolites of N-acetylated LKKTETQ were first identified from in vitro studies. First, the parent peptide and its metabolites were isolated from equine urine or plasma by solid-phase extraction using ion-exchange cartridges and then analyzed by liquid chromatography-mass spectrometry (LC/MS). Subsequently, these analytes were identified according to their LC retention times and the relative abundances of the major product ions. Notably, the peptide N-acetylated LKKTETQ could be detected and confirmed at concentrations as low as 0.02 ng/mL in equine plasma and 0.01 ng/mL in equine urine. Moreover, this method was successful in confirming the presence of N-acetylated LKKTETQ and its metabolites in equine urine and plasma collected from horses administered a single dose of TB-500 (containing 10 mg of N-acetylated LKKTETQ). Importantly, to our knowledge, this is the first identification of its metabolites in post-administration samples from horses.
Research Highlights
A product called TB-500, claimed to increase muscle growth and tissue repair in horses and other mammals, is available on the internet and officially distributed. It is presented as “the synthetic peptide of the active region of thymosin beta 4 (Tβ4)”, without any further qualitative description such as amino acid sequence or molecular weight. Tβ4, a 43 amino acid peptide, is the most abundant member of beta thymosins. Its primary function is to stimulate the production of T cells, which are an important part of the immune system. Additionally, Tβ4 has been shown to prevent apoptosis and promote cell survival, angiogenesis, and tissue regeneration in mice and rats.
